活性氧(Reactiveoxygenspecies,ROS)主要包括羥基自由基、超氧自由基和過氧化氫。在細胞或組織的正常生理代謝過程中會產生活性氧,同時一些環境因子例如紫外照射、吸煙、環境污染等也可以誘導活性氧的產生。活性氧在體內的失衡,將會導致細胞脂、蛋白和DNA的氧化損傷,繼而導致各種腫瘤、風濕性關節炎、糖尿病、肝損傷、以及中樞神經系統疾病等。
當然,機體中也存在許多種抗氧化的物質,包括抗氧化大分子、抗氧化小分子和酶等,能夠清除體內產生的各種活性氧,以阻止活性氧造成的氧化損傷。一個體系內的各種抗氧化大分子、抗氧化小分子和酶的總的水平即體現了該體系內的總抗氧化能力。因此測定樣本中的總抗氧化能力(TAC)具有非常重要的生物學意義。

| 實驗原理 | 總抗氧化能力(TAC)檢測試劑盒通過單電子轉移(SET)機制測量來自各種樣品的生物分子的總抗氧化能力。TAC檢測基于抗氧化劑(如尿酸)將銅(II)還原為銅(I)。還原后,銅(I)離子進一步與偶聯顯色試劑反應,產生在490nm處具有最大吸光度的顏色??寡趸瘎┑膬粑舛戎蹬c已知的尿酸標準曲線進行比較。吸光度值與樣品的總還原能力成正比。結果表示為“μM銅還原當量”或“mM尿酸當量”。銅比基于鐵的抗氧化劑測定更有優勢,因為所有類別的抗氧化劑,包括硫醇,都可以在邊緣自由基干擾下進行檢測。此外,銅基反應的動力學也比鐵基反應快,這使得TACAssay成為高效準確估計還原或抗氧化能力的理想工具 |

1.總抗氧化能力(TAC)檢測試劑盒,有物種種屬限制嗎?
我們的總抗氧化能力(TAC)檢測試劑盒,適用于任何生物蛋白質樣品。因為試劑盒原理并不依賴于可能因物種而異的蛋白質結構。
2.是否可以檢測凍存過的樣本?
我們的總抗氧化能力(TAC)檢測試劑盒,通常適用于在 -80?C 下保存兩個月以內樣品,但我們建議盡可能測試新鮮樣品以獲得最佳結果。
3.在制備食品樣品時,1:2, w/v 是什么意思?
食品樣品應準備為 1 g 樣品:2 mL 去離子水。
4.檸檬酸鹽血漿樣品可以用于該測定嗎?
檸檬酸鹽血漿樣品與總抗氧化能力測定兼容。 但應避免使用EDTA,因為這會干擾檢測。
5.該試劑盒可以測量細胞培養上清液的總抗氧化能力嗎?
不幸的是,我們的總抗氧化能力測定與細胞培養上清液不兼容。該測定基于銅的還原,介質中存在的鐵會干擾該反應。
