艾美捷PNA Bio-Cy3標記的Cas9蛋白：

左圖：將不同量的Cas9蛋白與500ngsgRNA孵育。通過脂質體胺將RNP復合物轉染到NIH3T3細胞中。24小時后，從每種條件下制備DNA用錯配敏感核酸酶法（T7E1）檢測Cas9蛋白和sgRNA的活性。

右圖：對于體內成像，通過以下方法將500ng的Cas9蛋白和500ng的sgRNA轉染到NIH3T3細胞lipofectamine。24小時后，通過共焦顯微鏡對細胞進行再成像顯微鏡。

Cy3-Cas9蛋白的活性通過電穿孔進入細胞，然后進行T7E1測定來確認。熒光通過共聚焦顯微鏡確認。
