Cytoskeleton 纖維連接蛋白 HiLyte488 （牛血漿）的純度測定

纖連蛋白從牛血漿中純化。蛋白質純度通過在4-20%聚丙烯酰胺凝膠上掃描考馬斯藍染色蛋白質的密度測定來確定。HiLyte Fluor 488標記的纖連蛋白純度為>80%（圖1）。 

該蛋白被修飾以含有隨機表面賴氨酸共價連接的HiLyte Fluor 488（2）。熒光染料的活化酯用于標記蛋白質。標記化學計量是通過蛋白質和染料濃度的光譜測量來確定的。最終標記化學計量是每個蛋白質分子 1-3 種染料（圖 2）。

HiLyte Fluor 488標記的纖連蛋白可以使用350-450nm激發和500-550nm發射的濾光片組進行檢測。 纖連蛋白在SDS-PAGE上作為單個亞基運行，表觀分子量為230 kDa。FNR02以橙色凍干粉的形式提供。每瓶FNR02含有20μg蛋白質。
