DV4-E88單克隆抗體與登革熱病毒血清型4(DENV-4)E蛋白上的DIII結構域反應。這種血清型特異性抗體不會與登革病毒1型、登革病毒2型或登革病毒3型(即登革病毒的其他血清型)發生交叉反應。登革病毒是一種由埃及伊蚊和白紋伊蚊傳播的黃病毒,每年在熱帶和亞熱帶地區造成超過1億人感染。輕度登革熱的特征是發燒、頭痛和肌痛,而重度登革熱可導致出血和毛細血管滲漏綜合征,即登革熱出血熱(DHF)/登革熱休克綜合征(DSS)。從結構上講,DENV是一種單鏈正極性RNA病毒,其包膜(E)蛋白有三個結構域。DV4-E88單克隆抗體(構象地)結合DENV-4(E二聚體上的四表位)上結構域III(DIII)的側脊(LR)。該抗體不識別酵母表面表達或分離的DENV-4 E蛋白DIII。DIII-LR區含有宿主細胞表面受體識別位點,參與病毒中和抗體的誘導。在體外研究中,DV4-E88單克隆抗體中和了1036株、TVP-376株和TVP-986株DENV-4基因型II毒株。在涉及AG129小鼠的實驗中,在病毒感染前單次體內注射DV4-E88單克隆抗體(100-500?g/小鼠)對DENV-4毒株H-241和TVP-376提供了適度到顯著的保護。
