TNF-α（腫瘤壞死因子-α）被稱為惡病質，是一種由156個氨基酸組成的糖蛋白，由金屬蛋白酶作用加工的前體產生。1975年由卡斯韋爾首次分離，它是先天免疫的細胞因子。 白細胞介素-1 或細菌內毒素刺激免疫細胞（活化的巨噬細胞、NK 細胞、單核細胞）和非免疫細胞（角質形成細胞、血管平滑肌細胞和顆粒葉黃素細胞）產生 TNF-α。 TNF-α的主要功能是作為免疫調節劑和宿主抵抗感染現象中炎癥反應的介質。它允許中性粒細胞和巨噬細胞募集到感染部位，通過刺激內皮細胞產生粘附分子和趨化因子（趨化細胞因子）。TNFalpha也作用于下丘腦以引發發燒。 TNF-α 的過度產生與許多病理狀況有關，包括惡病質、感染性休克和自身免疫性疾病。
