檢測原理：

雙抗體夾心ELISA的原理如圖1所示。在這種測定中，樣品中存在的IgG與抗IgG抗體反應，抗IgG抗體已吸附在聚苯乙烯微量滴定池的表面。在通過洗滌去除未結合的蛋白質后，加入與辣根過氧化物酶（HRP）綴合的抗IgG抗體。這些酶標記的抗體與先前結合的IgG形成復合物。在另一個洗滌步驟之后，通過添加無色底物3，3’，5，5’-四甲基聯苯胺（TMB）來測定與免疫吸附結合的酶。結合酶的數量與所測試樣品中IgG的濃度直接相關；因此，在450nm處的吸光度是測試樣品中IgG濃度的測量值。測試樣品中IgG的量可以根據標準曲線進行插值，并根據樣品稀釋度進行校正。
