檢測原理：

雙抗體夾心ELISA的原理在圖1中表示。在這個檢測中，樣本中存在的IgE與已經吸附在聚苯乙烯微孔板表面的抗IgE抗體發生反應。通過洗滌去除未結合的蛋白質后，加入與辣根過氧化物酶（HRP）偶聯的抗IgE抗體。這些酶標記的抗體與之前結合的IgE形成復合物。在另一個洗滌步驟之后，通過添加一種無色底物，四甲基聯苯胺（TMB），來檢測免疫吸附劑上結合的酶。結合酶的數量與測試樣本中IgE的濃度直接相關；因此，450納米處的吸光度是測試樣本中IgE濃度的量度。測試樣本中的IgE數量可以通過從標準品構建的標準曲線插值得到，并根據樣本稀釋進行校正。
