1994年人們首次發現了瘦素（leptin,LEP），瘦素是一個由脂肪組織分泌的蛋白類激素，而指導其合成的基因被稱為肥胖基因（obesegene，ob基因）。瘦素在血液中是分子質量為16kDa的具有親水性的單體，它是167個氨基酸在入血時自行去除掉N端21個氨基酸信號肽后所形成。小鼠與人的瘦素受體有將近80%的同源性。經研究短型LRa和長型LRb是發揮主要作用的兩種受體。

瘦素和瘦素受體與肥胖的發生有著密切關系。瘦素受體在下丘腦中廣泛分布,瘦素可以幫助建立、調節下丘腦中多條調控代謝的神經通路,影響中樞神經系統對糖脂代謝平衡、能量代謝平衡的調控。
